Pepstatin ist ein potenter Inhibitor von Aspartyl-Proteasen. Es ist ein Hexa-Peptid, das die ungewöhnliche Aminosäure statine (Sta, (3S, 4S) -4-Amino-3-hydroxy-6-methylheptansäure enthält) mit der Sequenz Isovaleryl-Val-Val-Sta-Ala-Sta ( Iva-Val-Val-Sta-Ala-Sta). Es wurde ursprünglich aus Kulturen verschiedener Arten von Actinomyces isoliert, aufgrund seiner Fähigkeit, Pepsin bei pikomolaren Konzentrationen zu hemmen. Pepstatin A ist bekanntlich ein Inhibitor von Aspartat-Proteinasen wie Pepsin, Cathepsinen D und E. Außer seiner Rolle als Proteinase-Inhibitor ist jedoch die pharmakologische Wirkung von Pepstatin A auf Zellen unklar. Pepstatin A unterdrückt den Rezeptoraktivator der NF-κB-Liganden (RANKL) -induzierten Osteoklasten-Differenzierung. Pepstatin A unterdrückt die Bildung von mehrkernigen Osteoklasten dosisabhängig. Diese Hemmung der Bildung betraf nur Osteoklastenzellen, dh nicht osteoblastenartige Zellen. Darüber hinaus unterdrückt Pepstatin A auch die Differenzierung von Vor-Osteoklasten-Zellen zu mononuklearen Osteoklastenzellen dosisabhängig. Diese Hemmung scheint unabhängig von den Aktivitäten von Proteinasen wie Kathepsin D zu sein, da die Bildung von Osteoklasten nicht mit der Konzentration unterdrückt wurde, die die Aktivität von Cathepsin D hemmte. Die Zell-Signalisierungsanalyse zeigte, dass die Phosphorylierung von ERK in Pepstatin A- Behandelten Zellen, während die Phosphorylierung von IκB und Akt fast keine Veränderung zeigte. Weiterhin verringerte Pepstatin A die Expression des Kernfaktors der aktivierten T-Zellen c1 (NFATc1). Diese Ergebnisse deuten darauf hin, dass Pepstatin A die Differenzierung von Osteoklasten durch die Blockade der ERK-Signalisierung und die Hemmung der NFATc1-Expression unterdrückt.

Pepstatin ist in Wasser, Chloroform, Ether und Benzol praktisch unlöslich, kann aber in Methanol, Ethanol und DMSO mit Essigsäure auf zwischen 1 und 5 mg / ml gelöst werden.
